- During the pause, don't image (i.e. no laser)
- Repeat (e.g. 10 times)

- Define the imaging
- In Slidebook, click on the
Home
tab, then hit Capture
- Define the parameters of the capture
- Exposure time, # of timepoints, interval, camera, etc
- When happy, go to the Capture Settings pull down in the top, left of the Capture GUI
- Define the TTL pulse
- Plug in Grass Stimulator to
TRIG/IN1
on the back of the TTL box.
- Push the Advanced button below Capture Settings in the GUI
- Under the General tab, and then within Captured Image Updates, select When Possible
- Under the Synchronization tab, define your TTL pulse
- Define when/how often in the top portion
- Define TTL pulse duration in the bottom portion
- Hit OK
- Define the pause
- The trick to defining the pause is to set up a dummy capture
- 3i's Mike Muntifering set up an Orca Flash dummy setting called Flash Dummy
- Flash Dummy can be selected from the Filter Set pull down within the Capture GUI
- Select Flash Dummy and define a fake exposure time and # of points that will collectively achieve the total duration of your desired pause
- Save the dummy capture settings in the Capture Settings pull down (upper left of Capture)
- Define the repeat
- Under the Special Capture tab, select Rapid 2D Capture
- Select the 6D tab
- Add the Capture script within the Capture Script pull down menu (top middle)
- Hit the Set button on the left top-ish
- Add the Pause script within the same Capture Script pull down menu
- Under the Special Capture tab, select 6D Multicapture
- Within the GUI, you should see the two scripts you selected above
- Select Sequential within the Capture Type pull down (upper left)
- Select the number of times you want to repeat within Repeat box
- Hit Start
